Sample Acquisition
Fully assisted sample acquisition is provided during regular staffed hours. Post-data processing (normalization and debarcoding) are included for all reservations. Data will be shared through Partners' Secure File Share within 24 hours from the end of the reservation. |

Final Sample Preparation
(For shipped samples only) Core staff will perform the final washes (twice with Cell Staining Buffer (CSB) and twice with Cell Acquisition Solution PLUS (CAS+). They will then perform a cell count, pellet, and acquire on the CyTOF-XT instrument. |

- Rates are calculated in 0.5 hour increments.
- 1 hour minimum reservation for instrument use.

Sample Requirements
- Must contain viability marker (103Rh, 194Pt, cisPt, or 198Pt).
- Must be FIXED with minimum 1.6% PFA.
- Metal Free BSA (Sigma)
- Must contain Ir191/193 for cell identification.
- Must be brought in 5mL polypropylene tubes
- MAXIMUM of 1 million pelleted cells per tube
- Individual or barcoded samples that exceed 1 million cells should be split evenly across multiple tubes (for example, a sample with 3 million cells should be split into 3 polypropylene tubes with 1 million cells in each tube).
- Cell Counts (including Cell Viability) obtained prior to final pelleting, must be provided to staff
- Always perform 2 final washes using CAS Plus.
- Samples are added to the Autosampler carousel in Pellet Form ONLY
- DO NOT ADD BEADS (Autosampler adds EQ6 beads and resuspends the sample in CAS Plus prior to acquisition on-board the instrument.)

Shipped Samples
The CyTOF core accepts shipped samples!

- Samples must be stained by the researcher.
- Shipped overnight with ice packs as a pellet in residual volume FOLLOWING DNA intercalator and fix / perm washing steps.
- The package must contain a TYPED list of the samples with cell counts (including viability) and a copy of the antibody panel.
- Reservations for shipped samples should be made no earlier than 12:00pm on the day the sample is scheduled to arrive.
- Researchers should give the Core staff the tracking number for the shipment.
- Core staff will perform final washes in CAS+, counting and resuspension/ filtering steps one hour prior to the scheduled reservation time.
- Technician time for final sample prep is charged at the sample preparation rate - 1 hour for shipped samples
